RESUMO

As one of the three major urban agglomerations in China, the Beijing-Tianjin-Hebei Region has strong economic strength but its ecological fragility is very prominent. To pursue the comprehensive development of economy and ecology, it is very important to analyze the ecological environment in the Beijing-Tianjin-Hebei Region. Here, the Beijing-Tianjin-Hebei Region was selected as the research area, and 19 indicators were selected to construct an evolution system based on the PSRM model. The temporal and spatial evolution characteristics of ecological vulnerability in the Beijing-Tianjin-Hebei Region were explored by combining the order relation method, CRITIC method, Theil index, and hot spot analysis, and the influencing factors were calculated via geographic detector. The results showed that:① the ecological vulnerability first increased and then decreased in the Beijing-Tianjin-Hebei Region. The vulnerable areas showed a northeast-southwest trend, and the ecological environment in the northeast and southwest regions was better than that in the central and southern regions. The area of slight vulnerability in 2014 increased by 6803.01 km2 compared with that in 2009. The area of mild vulnerability decreased by 130.41 km2, and the area of moderate vulnerability decreased by 26537.31 km2 compared with that in 2009. The areas of severe and extremely vulnerable status increased by 19512.9 km2 and 351.81 km2, respectively, compared with those in 2009. The habitat situation in the Beijing-Tianjin-Hebei Region improved significantly from 2014 to 2019. Compared with that in 2014, the areas of mild, moderate, severe, and extremely vulnerable decreased by 2248.29 km2, 2220.21 km2, 7988.67 km2, and 55.98 km2, respectively. The light area increased by 12513.15 km2 compared with that in 2014. ② According to the calculation results of the Theil index, the spatial correlation degree of ecological vulnerability in the Beijing-Tianjin-Hebei Region exhibited a V-shaped fluctuation, and the spatial pattern of the cold and hot areas was predominantly consistent with that of the vulnerability. ③ Biological abundance, PM10, and the human disturbance index had a significant influence on the spatial differentiation of ecological vulnerability in the Beijing-Tianjin-Hebei Region. Based on the results of ecological vulnerability analysis, some suggestions on the ecological environment and sustainable development in the Beijing-Tianjin-Hebei Region were proposed.

Behaviors emerge via a combination of experience and innate predispositions. As the brain matures, it undergoes major changes in cellular, network, and functional properties that can be due to sensory experience as well as developmental processes. In normal birdsong learning, neural sequences emerge to control song syllables learned from a tutor. Here, we disambiguate the role of tutor experience and development in neural sequence formation by delaying exposure to a tutor. Using functional calcium imaging, we observe neural sequences in the absence of tutoring, demonstrating that tutor experience is not necessary for the formation of sequences. However, after exposure to a tutor, pre-existing sequences can become tightly associated with new song syllables. Since we delayed tutoring, only half our birds learned new syllables following tutor exposure. The birds that failed to learn were the birds in which pre-tutoring neural sequences were most 'crystallized,' that is, already tightly associated with their (untutored) song.

Renal fibrosis underlies all forms of end-stage kidney disease. Endophilin A2 (EndoA2) plays a role in nephrotic syndrome; however, its effect on renal fibrosis remains unknown. Here, we demonstrate that EndoA2 protects against kidney interstitial fibrosis via the transforming growth factor-ß (TGF-ß)/Smad signaling pathway. Mouse kidneys with fibrosis or kidney biopsy specimens from patients with fibrotic nephropathy had lower levels of EndoA2 protein expression than that in kidneys without fibrosis. In vivo overexpression of EndoA2 with the endophilin A2 transgene (EndoA2Tg ) notably prevented renal fibrosis, decreased the protein expression of profibrotic molecules, suppressed tubular injury, and reduced apoptotic tubular cells in the obstructed kidney cortex of mice with unilateral ureteral obstruction (UUO). In vivo and in vitro overexpression of EndoA2 markedly inhibited UUO- or TGF-ß1-induced phosphorylation of Smad2/3 and tubular epithelial cells dedifferentiation. Furthermore, EndoA2 was co-immunoprecipitated with the type II TGF-ß receptor (TßRII), thus inhibiting the binding of the type I TGF-ß receptor (TßRI) to TßRII. These findings indicate that EndoA2 mitigates renal fibrosis, at least partially, via modulating the TGF-ß/Smad signaling. Targeting EndoA2 may be a new potential therapeutic strategy for treatment of renal fibrosis.

BACKGROUND: Circular RNA (circRNA) is involved in the process of acute kidney injury (AKI), but only a few circRNAs have been reported. In the study, we investigated a new circRNA and its association with AKI. METHODS: An AKI model was established in Sprague-Dawley rats, followed by serum creatinine and urea nitrogen tests measured by a biochemical analyzer. The pathological changes and apoptosis in the renal tissue were detected by Hematoxylin and Eosin, and TUNEL staining. Then, circRNA expression in AKI was determined by quantitative real-time-PCR (qRT-PCR). NRK-52E cells were induced with hypoxia/reoxygenation (H/R) as in vitro models and the circ-Snrk level was tested by qRT-PCR. The effects of circ-Snrk in H/R-induced NRK-52E cells were assessed by flow cytometry, western blot, and enzyme-linked immunosorbent assay. Finally, RNA sequencing and western blot analysis were used to validate the mRNA profile and pathways involved in circ-Snrk knockdown in H/R-induced NRK-52E. RESULTS: A reliable AKI rat model and H/R cell model were established. qRT-PCR demonstrated that circ-Snrk level was upregulated in AKI left kidney tissue and NRK-52E cells with H/R treatment. Circ-Snrk knockdown inhibited apoptosis of NRK-52E cells and secretion of inflammatory factors (IL-6 and TNF-α). RNA sequencing showed that the mRNA profile changed after inhibition of circ-Snrk and differential expression of mRNA mainly enriched various signaling pathways, including MAPK signaling pathway. Furthermore, western blot indicated that circ-Snrk knockdown could inhibit the activation of p-JNK and p-38 transcription factors. CONCLUSIONS: Circ-Snrk is involved in AKI development and associated with the MAPK signaling pathway in AKI.

Carbon dioxide (CO2) emissions from inland waters to the atmosphere are a pivotal component of the global carbon budget. Anthropogenic land use can influence riverine CO2 emissions, but empirical data exploring cause-effect relationships remain limited. Here, we investigated CO2 partial pressures (pCO2) and degassing in a monsoonal river (Yue River) within the Han River draining to the Yangtze in China. Almost 90% of river samples were supersaturated in CO2 with a mean ± standard deviation of 1474 ± 1614 µatm, leading to emissions of 557 - 971 mmol/m2/day from river water to the atmosphere. Annual CO2 emissions were 1.6 - 2.8 times greater than the longitudinal exports of riverine dissolved inorganic and organic carbon. pCO2 was positively correlated to anthropogenic land use (urban and farmland), and negatively correlated to forest cover. pCO2 also had significant and positive relationships with total dissolved nitrogen and total dissolved phosphorus. Stepwise multiple regression models were developed to predict pCO2. Farmland and urban land released nutrients and organic matter to the river system, driving riverine pCO2 enrichment due to enhanced respiration in these heterotrophic rivers. Overall, we show the crucial role of land use driving riverine pCO2, which should be considered in future large-scale estimates of CO2 emissions from streams. Land use change can thus modify the carbon balance of urban-river systems by enhancing river emissions, and reforestation helps carbon neutral in rivers.

Headwater streams make the majority of cumulative stream length in a river basin, carbon dioxide (CO2) emission from headwater (low order) streams is thus an essential component. Anthropogenic activities in headwater areas such as land use change and land use practices can strongly modify terrestrial carbon and nutrient input, which could affect the level of partial pressure of dissolved carbon dioxide (pCO2) and CO2 degassing from streams. However, there are large uncertainties in estimates due to the lack of data in subtropical rivers of rapidly developing rural regions. The spatiotemporal variation and driving factors of the pCO2 and CO2 degassing from low-order streams remain to be explored. In this study, we assess multi-spatial scale effects of land use on pCO2 dynamics in seven headwater tributary rivers in Central China during 2016, 2017 and 2018 in rainy and dry seasons. Our results reveal that the stream pCO2 level consistently increases as the stream order increases from 1 to 3 under apparent seasonal variations. Riverine pCO2 is positively related to the percentage of urban land and cropland surrounding the river segments, but is negatively related to the percentage of forest land. The stream pCO2 is more closely correlated with the 1000 and 2000 m diameters of circular buffers at upstream sampling sites than the circular buffers with 100 and 500 m diameters. There exist significant relationships of pCO2 with the concentrations of TN, TP, DO, and DOC in the low-order streams. The partial redundancy analysis quantifies the relative importance of anthropogenic land uses, natural factors and water chemical variables in mediating stream pCO2, showing that influences of anthropogenic land uses (urban and cropland) on pCO2 decrease, with a percentage role of 34%, 14%, and 4% in the 1st-, 2nd- and 3rd-order streams, respectively. The impact of nutrients on pCO2, however, increases as the stream order increases. Urban influence on stream pCO2 also decreases as stream order increases. Our study highlights the effect of land use/land cover types and stream order on riverine pCO2 and provides new insight into estimating CO2 emission in headwater streams. Future studies are needed on the linkage between riverine CO2 degassing and stream orders under changing land use conditions.

BACKGROUND: Broad hemodynamic changes, is believed to have a profoundly damaging effect on donor livers after brain death (DBD) or cardiac death (DCD). It remains unclear whether Doppler ultrasonography (DUS) and contrast-enhanced ultrasonography (CEUS), the imaging modalities to evaluate perfusion, could provide more information of liver discarded. OBJECTIVE: To evaluate the ability of DUS and CEUS to predict the risk of DBD or DCD liver discarded. METHODS: The consecutive DBD or DCD donors with DUS/CEUS examinations before surgical procurement from February 2016 to June 2018 at our institution were included. The US and CEUS images of each donor liver were analyzed and the parameters were recorded. RESULTS: Among the 67 eligible donor livers, 15 (22.4%) were discarded and 52 (77.6%) were used. The discarded livers showed prolonged SAT of hepatic artery (0.08s vs 0.06s, OR = 2.169, P = 0.008) on DUS, less cases with homogeneous enhancement (40.0% vs 73.1%, OR = 0.243, P = 0.028) on CEUS, more cases with decreased enhancement (53.3% vs 19.2%, OR = 4.800, P = 0.009), and less difference of the peak time between portal vein and liver parenchymal (0.5s vs 6.7s, OR = 0.917, P = 0.034). The multivariable analysis showed that donor liver with prolonged SAT of hepatic artery (OR = 7.304, 95% CI: 1.195-44.655, P = 0.031) and decreased enhancement (OR = 2.588, 95% CI: 1.234-5.426, P = 0.012) were independent factors of liver discarded. CONCLUSIONS: DUS/CEUS could be applied as a promising predictive tool to screen high-risk liver donors. The prolonged SAT of hepatic artery on DUS and the decrease of liver donor in enhancement on CEUS, indicating hemodynamic changes in DBD and DCD donor livers, were risk factors of liver discarded.

PURPOSE: To evaluate whether Doppler ultrasonography (DUS) and contrast-enhanced ultrasonography (CEUS) can identify liver donation after brain death (DBD) and cardiac death (DCD) with the risk of developing short-term primary graft dysfunction (PGD) or arterial and biliary complications within 1 year. MATERIALS AND METHODS: Consecutive DBD and DCD donors who underwent DUS/CEUS examinations before surgical procurement from February 2016 to June 2018 at our institution were included. The US and CEUS images of each donor liver were analysed, and the parameters were recorded. RESULTS: The mean time for US examination was 32 min (range, 20-59 min), and all donors tolerated the examination well. In terms of short-term outcomes, among the 52 eligible donor livers, 20 (38.5 %) of their recipients developed PGD. The multivariable analysis showed that decreased enhancement of donor livers on CEUS (OR = 15.976, 95 % CI: 1.652-154.628, P = 0.017) and high recipient model for end-stage liver disease (MELD) scores (OR = 1.050, 95 % CI: 1.004-1.099, P = 0.034) before liver transplantation (LT) were independent factors of PGD. In contrast, for long-term complications, among the 48 eligible donor livers, 16 (33.3 %) developed arterial or biliary complications within 1 year. The multivariable analysis did not show any independent factors of arterial or biliary complications within 1 year. CONCLUSIONS: A decrease in enhancement on CEUS is an independent risk factor for poor short-term outcomes of LT. CEUS may be promising for predicting post-LT outcomes of critically ill donors effectively and safely by evaluating the haemodynamic changes in DBD and DCD donor livers.

To investigate the mechanism of renal ischemia-reperfusion injury (IRI) via regulation of N6-methyl-adenosine (m6A) and relevant genes, IRI was induced in Sprague-Dawley rats, and urine and serum creatinine levels and tissue structure changes were observed. m6A and methyltransferase-like 3 (METTL3) protein levels were assessed via dot-blot and Western blot analyses, respectively. The hypoxia/reoxygenation (H/R) cell model was constructed using NRK-52E cells, and METTL3 protein levels were assessed. METTL3 was inhibited to observe its impact on NRK-52E cell apoptosis and m6A expression in H/R processes. Methylated RNA immunoprecipitation (MeRIP) sequencing was conducted followed by MeRIP-quantitative RT-PCR and quantitative RT-PCR validation. Our results indicated that urine and serum creatinine levels increased and that renal injury and cell apoptosis were both observed in the IRI model. In additon, m6A expression increased in the IRI model, and METTL3 protein levels significantly increased in the IRI and H/R models. When METTL3 was inhibited, m6A levels were accordingly decreased and cell apoptosis was suppressed in the H/R in vitro model. Based on MeRIP sequencing, transcription factor activating enhancer binding protein 2α (tfap2a), cytochrome P-450 1B1 (cyp1b1), and forkhead box D1 (foxd1) were significantly differentially expressed, as was m6A, which is involved in the negative regulation of cell proliferation and kidney development. We confirmed that foxd1 mRNA and its methylation levels contributed to IRI and H/R.

In this work, a dual-lithium salt was proposed for constructing an electrolyte for high energy density lithium ion batteries. LiPO2F2 was composed with traditional LiPF6 to enhance the high voltage performance of the electrolyte. The electrochemical performance of the NCM811/Li cells with LiPO2F2/LiPF6 dual-lithium salt at 2.8-4.5 V was investigated. It was found that the dual-lithium salt can inhibit the oxidative decomposition of the electrolyte, suppress the dissolution of the transition metal ions in the electrode material, and reduce the side reaction between the transition metal ions and the electrolyte. We believe that the strategy of a dual-lithium salt electrolyte may provide a new idea to stabilize the electrode/electrolyte interface at high voltage, which is very important for developing high energy density batteries.

Identifying low-dimensional features that describe large-scale neural recordings is a major challenge in neuroscience. Repeated temporal patterns (sequences) are thought to be a salient feature of neural dynamics, but are not succinctly captured by traditional dimensionality reduction techniques. Here, we describe a software toolbox-called seqNMF-with new methods for extracting informative, non-redundant, sequences from high-dimensional neural data, testing the significance of these extracted patterns, and assessing the prevalence of sequential structure in data. We test these methods on simulated data under multiple noise conditions, and on several real neural and behavioral datas. In hippocampal data, seqNMF identifies neural sequences that match those calculated manually by reference to behavioral events. In songbird data, seqNMF discovers neural sequences in untutored birds that lack stereotyped songs. Thus, by identifying temporal structure directly from neural data, seqNMF enables dissection of complex neural circuits without relying on temporal references from stimuli or behavioral outputs.

INTRODUCTION: Hepatocellular carcinoma (HCC) accounts for more than 90% of primary liver cancer. Although great progress has been made on HCC molecular mechanism and therapy techniques, the prognosis of HCC patient is poor due to high metastasis and recurrence. MATERIALS AND METHODS: Expression of miR-542-3p was quantified by quantitative real-time PCR (qRT-PCR). The role of miR-542-3p in HCC metastasis was examined using transwell and 3D-culture assay. qRT-PCR, Western blotting and luciferase reporter assay were used to elucidate the mechanisms of miR-542-3p-mediated cancer metastasis. RESULTS AND CONCLUSION: In the research, we found that miR-542-3p is decreased in HCC cell lines and tissues, and downregulation of miR-542-3p enhances, while upregulation suppresses HCC cell invasion ability. Further assay demonstrated that miR-542-3p can directly target TGF-ß1 3' untranslated region (3'UTR) to influence TGF-ß/Smad signaling pathway, and suppression of miR-542-3p can hyperactivate TGF-ß/Smad pathway and further to promote Epithelial-Mesenchyme Transition (EMT) and induce poor prognosis. Lastly, the clinical correlation analysis illustrated that miR-542-3p is negatively related with the activity of TGF-ß1. In summary, our results find that miR-542-3p takes an important role on HCC progression and provide more evidence of microRNAs (miRNAs) for cancer therapy.

We design and demonstrate a birefringent Michelson interferometer based interleaver with ultra-low dispersion and low cost. The interleaver consists of polarizing beam splitters (PBS's) and quarter-wave plates and half-wave plates. The PBS's based Michelson interferometers provide the optical path difference for interference between the two orthogonal polarization components and the half-wave plates provide the birefringent needed to minimize ripple of output. The designed interleaver with two-stage interferometer in a 50 GHz channel spacing application exhibits a 0.5 dB passband and a 25 dB stopband both 27 GHz; a channel isolation higher than 35 dB and chromatic dispersion less than ±5 ps/nm within 0.5 dB passband; 1.3 dB insertion loss and 0.3 dB PDL; 0.04 GHz/°C thermal stability. Since all of the optical components can be optically bonded together, the device is robust and easy to be aligned, which reduces labor cost.

We present a single longitudinal mode, diode pumped Nd:YVO(4) microchip laser where a pair of quarter-wave plates (QWPs) sandwich Nd:YVO(4) and the principle axes of QWPs are oriented at 45 degrees to the c-axis of Nd:YVO(4). Three pieces of crystals were optically bonded together as a microchip without adhesive. Owing to large birefringence of Nd:YVO(4), two standing waves with orthogonal polarizations compensate their hole-burning effects with each other, which diminish total spatial hole-burning effects in Nd:YVO(4). The maximum pump power of greater than 25 times the threshold for single longitudinal mode operation has been theoretically shown and experimentally demonstrated. The power of output, slope efficiencies and temperature range of single longitudinal mode operation are greater than 730 mw (at 1.25 W pump), 60% and 30 degrees C, respectively.
